An aqueous 0.5 M solution of lithium hydroxide (625 μL, 0.081 mmol) was added to a stirred room temperature solution of the product of Example 13 (11 mg, 0.026 mmol) in methanol (100 μL). The mixture was stirred for 16 hours at room temperature and was then concentrated in vacuo to a crude solid/liquid mixture. The residue was purified by mass triggered reverse phase HPLC. The product containing fractions were lyophilized to give the title compound (TFA salt) as a white solid. LRMS calc'd for C23H28N5O2 [M+H]+, 407; found 406. Jak1 activity: +.
