Procedure
The above-obtained 4-(4-dimethylaminophenyl)-2,4-dioxobutanoic acid methyl ester (742 mg) and 3-chloro-6-hydrazinopyridazine (473 mg) in methanol (30 mL) was refluxed under heat for 18 hours, and then cooled in air. The reaction solvent was removed under reduced pressure, and the residue was partitioned by use of saturated aqueous sodium hydrogencarbonate and chloroform. The organic layer was dried over sodium sulfate anhydrate, followed by filtration. The solvent was removed under reduced pressure, and the residue was purified through silica gel column chromatography (chloroform-methanol), to thereby give 1-(6-chloro-3-pyridazinyl)-5-(4-dimethylaminophenyl)pyrazole-3-carboxylic acid methyl ester as a solid (679 mg, 63%).
