Reaction Steps
A methanol solution (10.0 mL) containing N-{5-bromo-4-(4-(4-methoxyphenoxy)-2,6-dimethylphenyl)thiazol-2-yl}acetamide (2-3, 465 mg, 1.3 mmol) and 6 N hydrochloric acid (6.0 mL) was heated at reflux for 4.0 h. The solution was added with 10% aqueous NaOH and the resultant solids were collected to give 5-bromo-4-(4-(4-methoxyphenoxy)-2,6-dimethylphenyl)thiazol-2-amine (2-4, 325 mg) as white solids in 55% yield: 1H NMR (500 MHz, DMSO-d6) δ 6.97-7.04 (m, 4H), 6.65 (s, 2 H), 3.76 (s, 3 H), 2.03 (s, 6 H). ESI-MS: m/z 405.3, 407.2 (M+H)+.
