Reaction Steps

To a solution of trans-5-benzyloxyamino-2-cyano-piperidine-1-carboxylic acid-1-tert-butyl ester (6 gm, 0.018 mol) in toluene (120 ml) were added sodium azide (5.88 gm, 0.090 mol), triethylamine hydrochloride (12.42 gm, 0.090 mol) and the resulting mixture was heated to 100° C., under stirring for 2 hours. The reaction mixture was cooled to 25° C. to 35° C. and poured into saturated aqueous ammonium chloride solution (150 ml). The mixture was stirred for 15 min. and aqueous layer was extracted with ethyl acetate (1×120 ml). The combined organic layer was dried over sodium sulfate and was concentrated under vacuum to obtain the step-1 product as colorless oil, in 6.0 gm quantity (yield 89%).
