Reaction Steps

To a round bottom flask, a mixture of 1-methyl-2-(piperidine-3-yloxy)-1H-[4,4′]bipyrimidinyl-6-one (0.2 g, 0.70 mmol), bromobenzene (105 μl, 1.0 mmol), tris(dibenzylideneacetone)dipalladium(0) (46 mg, 0.05 mmol), 2-dicyclohexylphosphino-2′,6′-dimethoxybiphenyl (82 mg, 0.2 mmol) and potassium phosphate tribasic (0.43 g, 2.0 mmol) in dimethoxyethane (3 ml) was added at room temperature under nitrogen atmosphere and heated to 100° C. After stirring for 5 hours, the resulting mixture was cooled to room temperature. The resulting mixture was poured into water and extracted with chloroform. The organic layer was dried over anhydrous sodium sulfate and concentrated in vacuo. The residue was purified by silica gel column chromatography (eluent; hexane/ethyl acetate=50/50) to afford 1-methyl-2-(1-phenyl-piperidin-3-yloxy)-1H-[4,4′]bipyrimidinyl-6-one as a yellow solid (67 mg, 0.18 mmol, 26%).
