Reaction Steps

To a solution of methyl 2,4-bis(benzyloxy)-5-ethyl-6-(1-methyl-1H-indol-5-yl)nicotinate (880 mg, 1.74 mmol) in DMF (10 mL) was added chlorosulfonyl isocyanate (0.18 mL, 2.07 mmol, 1.2 eq) at 0° C. The reaction was monitored by LC-MS and starting material was completely consumed within 5 min. The reaction was quenched with water then extracted by CH2Cl2 (3×30 mL). The combined organic layers were dried over Na2SO4 and then concentrated to give a crude product which was purified by trituration with hexanes and CH2Cl2 to afford the title compound (732 mg, 79%).
