Procedure

Compound 2 (3.6 g, 18.9 mmol) and (R)-1-Boc-3-aminopiperidine (9.45 g, 47.2 mmol) were dissolved in ethanol (50 mL) and TEA (10.5 mL, 76 mmol) was added. The reaction was sealed and heated to 130° C. for 72 h. The orange solution was concentrated and the residue was purified by column chromatography on silica gel, eluting with EtOAc/hexane to give the title compound as a white foam. 1H NMR (CDCl3) δ 5.40 (s, 1H), 4.78 (br s, 1H), 3.88 (s, 3H), 3.84-3.81 (m, 1H), 3.58-3.56 (m, 2H), 3.17-3.05 (m, 2H), 2.49 (s, 3H), 2.04-1.91 (m, 1H), 1.75-1.67 (m, 1H), 1.57-1.51 (m, 2H), 1.44 (s, 9H); LCMS 2.30 min, 355 (M+H).
