Reaction Steps
To a solution of 1-methyl-4-{[(6-oxo-7,8,9,10-tetrahydro-6H-pyrido[3′,2′:4,5]pyrrolo[1,2-a][1,4]diazepin-2-yl)carbonyl]amino}-1H-imidazole-2-carboxylic acid (50 mg, 0.14 mmol) in DMF (1.5 mL) was added cyclopentylamine (27 μL, 0.27 mmol), HOBt (18 mg, 0.14 mmol), N,N-diisopropylethylamine (47 μL, 0.27 mmol), and DMAP (3 mg, 0.027 mmol) followed by 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide hydrochloride (78 mg, 0.41 mmol). The mixture is stirred at room temperature for 64 h. The mixture is poured over H2O and the resulting solid is collected by filtration and is purified via preparative HPLC using a gradient elution from 10-75% acetonitrile/H2O with 0.1% TFA to obtain the title compound (6 mg, 10%). LCMS: 436.20 (M+H+). (System V-med polar)
