A mixture of crude 1-ethyl-2-{[2-(2,5-difluorophenyl)-1H-imidazol-1-yl]methyl}-5-azido-1H-benzimidazole (143 mg) and ethyl propiolate (1 eq.) in ethanol (10 mL) is heated at reflux for 6 h. The mixture is cooled, concentrated, and the residue purified by preparative silica gel chromatography to give 1-{2-[2-(2,5-Difluoro-phenyl)-imidazol-1-ylmethyl]-1-ethyl-1H-benzoimidazol-5-yl}-1H-[1,2,3]triazole-4-carboxylic acid ethyl ester. 1H NMR (CDCl3): δ 8.51s, (1H), 8.05 (s, 1H), 7.74 (m, 1H), 7.50-7.39 (m, 4H), 7.21-7.17 (m, 2H), 7.06 (s, 1H), 5.54 (s, 2H), 4.47 (q, 2H), 3.86 (q, 2H), 1.43 (t, 3H), 1.04 (t, 3H), LCMS MH+460.6.
