Reaction Steps

A mixture of 4-hydroxypyridine (10 g, 105 mmol), ethyl 4-bromobutyrate 12-1 (15.0 ml, 105 mmol) and Cs2CO3 (34.2 g, 105 mmol) in DMF (100 ml) was stirred at room temperature for 24 h. The reaction was filtered and the filtrate diluted with ethyl acetate (300 ml) and washed with water (4×100 ml) and brine (100 ml) then dried (Na2SO4), filtered, and evaporated. The resulting oil was purified by chromatography on silica gel (3% CH3OH/CH2Cl2) to give 12-2 as a colorless glass.
