Reaction Steps

To a solution of 6-fluoro-2-pyridinecarboxylic acid (15.0 g, 106.3 mmol) in tetrahydrofuran (200 ml) was added 1,1′-carbonylbis-1H-imidazole (19.0 g, 116.9 mmol), and the mixture was heated under reflux for 30 min. The reaction solution was cooled and monoethyl malonate magnesium salt (16.8 g, 58.5 mmol) was added. The mixture was stirred at room temperature for 30 min. 1N Hydrochloric acid (200 ml) was added to the reaction solution and the mixture was extracted with ethyl acetate (200 ml×2). The extract was washed with saturated brine, dried over anhydrous magnesium sulfate and evaporated under reduced pressure. The residue was purified by silica gel column chromatography (hexane:ethyl acetate=4:1) to give ethyl 3-(6-fluoro-2-pyridyl)-3-oxopropionate (20.16 g, 90%) as a brown oil.
