To a solution of (2-ethylamino-7-fluoro-6-methoxyquinolin-3-yl)methanol SMA 44090 (278 mg, 1.11 mmol) in dry CH2Cl2 (30 mL) at 0° C. under N2 in a 100 mL round-bottomed flask equipped with a magnetic stirrer was added dropwise SOCl2 (1.60 mL, 22.1 mmol) and the mixture was stirred for 3 h at RT then concentrated to dryness at 40° C. under vacuum. The residue was then diluted with CH2Cl2 (30 mL) and concentrated back to dryness at 40° C. under vacuum (done 3 times) to give 3-(chloromethyl)-N-ethyl-7-fluoro-6-methoxyquinolin-2-amine hydrochloride SMA 44096 as a yellow solid (348 mg, quantitative yield).
