Reaction Steps
To a solution of bis(pinacolato)diboron (94 mg,0.327 mmol), 5-Bromo-2-(4-fluoro-3-methyl-phenyl)-indole-1-carboxylic acid tert-butyl ester (75 mg, 0.18 mmol), and potassium acetate (55 mg, 55 mmol) in anhydrous 1,4-dioxane (250 uL) is added [1,1′-bis(diphenylphosphino)ferrocene]dichloropalladium(II) complex with dichloromethane (1:1) (PdCl2(dppf)*CH2Cl2) (45 mg, 0.06 mmol). The reaction mixture is heated under reflux at 100° C. for 2 hours under nitrogen. Under nitrogen, 4-{[(5-Bromo-1-methyl-1H-pyrazole-3-carbonyl)-amino]-methyl}-benzoic acid is added in N,N dimethylformamide (1 ml), followed by aqueous Na2CO3 (1 ml, 2N) and additional catalyst (20 mg). The reaction mixture is heated at 85° C. for 8 hours. The reaction mixture is diluted with 20 ml ethyl acetate, passed through a plug of celite and washed with water. The aqueous layer is evaporated, taken up in TFA/DCM (1:1, 5 ml) and stirred for 30 minutes. The solvent is evaporated in vacuo and the residue purified on reverse-phase HPLC to give the title compound 20 mg, (48%) LC/MS ESI m/z (M+H)+=484.5.
