A mixture of 4-(chloromethyl)-1-(3′-methylsulfonyl)biphenyl-4-yl)-2-(2-(trifluoromethyl)phenyl-1H-imidazole (1.1 g), NaCN (1.2 g, 24.5 mmol), and dry DMF was stirred at 100° C. for 12 h. The solvent was removed in vacuo, and the residue was partitioned between water and EtOAc, the phases were separated, and the aqueous phase was extracted with EtOAc. The combined extracts were washed with water, brine, dried over sodium sulfate, and evaporated in vacuo. The crude product was purified by flash chromatography (SiO2, 80% EtOAc/hexanes) to give 1-(1-(3′-methylsulfonyl)biphenyl-4-yl)-2-(2-(trifluoromethyl)phenyl-1H-imidazole-4-carbonitrile (730 mg, 74%) as a white solid. MS (ESI) 482 [M+H]+.
