Reaction Steps
To a solution of [2-(3-Cyclopentylamino-phenyl)-5-(2-trimethylsilanyl-ethoxymethyl)-5H-pyrrolo[2,3-b]pyrazin-7-yl]-(1-methyl-cyclohexyl)-methanone in dimethyl formamide was added sodium hydride (60% dispersion in mineral oil, 9 mg, 0.218 mmol) under argon. The reaction mixture was stirred at room temperature for 15 minutes before the addition of methyl iodide via syringe. The reaction was stirred for 14 h before quenching with water and extracting 3 times with dichloromethane. The combined organics were washed with water, dried over MgSO4, filtered and concentrated in vacuo. Preparative thin layer chromatography (25% EtOAc/hexanes) afforded 0.0152 g (15% yield) of [2-[3-(Cyclopentyl-methyl-amino)-phenyl]-5-(2-trimethylsilanyl-ethoxymethyl)-5H-pyrrolo[2,3-b]pyrazin-7-yl]-(1-methyl-cyclohexyl)-methanone as a yellow oil.
