Reaction Steps

The material contained only 22-25% of AT as determined by RID (using Meloy kit) and assuming E1% =5. AT (15.4 mg) was dissolved in 1.2 ml of 0.1 M NaHCO3 and labeled at pH 9.0 with 0.5 mg FITC as previously described. The resultant conjugate after separation on Sephadex G-25, was in 1.8 ml; OD280 =9.8, OD496 =15.6 (for fluorescein). AT was estimated by RID at 2.3-2.4 mg/ml.
