Reaction Steps
(R)-1-(2-methylpiperazin-1-yl)-4-phenylphthalazine (JK-10) (200 mg, 0.65 mmol) was dissolved in 5 mL of dichloromethane at room temperature. To this solution was added triethylamine (0.18 mL, 1.3 mmol, 2 eq) and benzoyl chloride (0.083 mL, 0.72 mmol, 1.1 eq). After 5 minutes, the reaction was diluted with 75 mL of ethyl acetate and washed with 5 mL of sat. NaHCO3, 5 mL of water, and 5 mL of brine. The organic phase was dried over MgSO4, filtered, and concentrated. Purification by column chromatography (35% ethyl acetate in hexanes) afforded 200 mg (75% yield) of (R)-(3-methyl-4-(4-phenylphthalazin-1-yl)piperazin-1-yl)(phenyl)methanone (4). MS (M+H)+=409.1
