(E)-[2-(3-{4-[[tert-Butoxycarbonyl-(2-morpholin-4-yl-propyl)-amino]-(4-isopropyl-phenylcarbamoyl)-methyl]-phenyl}-acryloylamino)-phenyl]-carbamic acid tert-butyl ester (crude product from Example 3) was dissolved in 1.25M HCl/MeOH (2.9 mL) and stirred at room temperature overnight. The reaction was quenched slowly with solid sodium bicarbonate until the pH was 6-7. The mixture was diluted in acetonitrile with a small amount of dimethylsulfoxide, passed through a 40 μm pipette filter, and then purified by preparative HPLC to obtain 150 mg desired product (56% over two steps). MS: calc'd 556 (MH+), exp 556 (MH+).
