Procedure

To a solution of 4-(2-amino-4-methyl-phenylamino)-3-methoxy-piperidine-1-carboxylic acid ethyl ester (800 mg, 2.60 mmol) in acetic acid (5 mL) was added 1,1,1-triethoxy ethane (550 mg, 3.12 mmol). After heating at 60° C. for 30 minutes, the mixture was concentrated under reduced pressure and the residue was purified by flash chromatography (15/1 CH2Cl2/CH3OH) to 4-(2-ethyl-5-methyl-benzoimidazol-1-yl)-3-methoxy-piperidine-1-carboxylic acid ethyl ester (940 mg, 89%) as a white solid.
