Procedure

To a solution of 3-(6-Cyano-3-naphthalen-1-ylmethyl-2-oxo-2,3-dihydro-benzimidazol-1-yl)-propionic acid ethyl ester (20 mg, 0.05 mmol) in MeOH (5 mL) was added 2N NaOH (3 mL). The reaction mixture was stirred at room temperature for 16 h. When all the starting material was consumed, the reaction mixture was acidified with 1N HCl. The reaction mixture was extracted with CH2Cl2 (×2). The combined organic phase was dried over Na2SO4 and concentrated. The resulting residue was purified by silica gel prep TLC using 10:1 CH2Cl2:MeOH as an eluent to afford 4 mg (22%) of the desired product as an off-white solid. LCMS (ESMS): m/z 372.11 (M+H+).
