Reaction Steps

To a solution of 5-methyl-1-phenethyl-4-phenyl-imidazole-2-carbaldehyde (example 103B) (50 mg, 0.172 mmole) in CHCl3 (1 mL) is added 4-hydroxy-N-methylpiperidine (198 mg, 1.72 mmoles). Methanesulfonic acid is added dropwise until a purple colour developed. The reaction is completed (TLC control). A saturated solution of Na2CO3 is added to reach pH 10. The aqueous phase is extracted three times with CH2Cl2. The organic phase is dried over magnesium sulphate, filtered and the solvent is removed under reduced pressure. The residue is purified by preparative TLC over silica gel using (CH2Cl2:MeOH:NH4OH) (92.5:7.5:0.75) to give 1-methyl-4-(1-methyl-piperidin-4-yloxy)-2-phenyl-9,10-dihydro-4H-3,10a-diaza-benzo[f]azulene.
