Reaction Steps
A 100 mL round-bottomed flask equipped with a magnetic stirrer and a reflux condenser was charged with 10-[4-(5-bromo-1-methyl-6-oxo-1,6-dihydropyridin-3-yl)-3-(hydroxymethyl)pyridin-2-yl]-4,4-dimethyl-1,10-diazatricyclo[6.4.0.02,6]dodeca-2(6),7-dien-9-one 102b (497 mg, 1.0 mmol), 102a (312 mg, 1.5 mmol), Pd2(dba)3 (92 mg, 0.10 mmol), XantPhos (4,5-Bis(diphenylphosphino)-9,9-dimethylxanthene, CAS Reg. No. 161265-03-8, 116 mg, 0.20 mmol), Cs2CO3 (652 mg, 2.0 mmol), and DMF (20 mL). After three cycles of vacuum/argon flush, the mixture was heated at 80° C. for 4 h. After this time the reaction was cooled to room temperature. It was then filtered and the filtrate was evaporated under reduced pressure. The residue was purified by reverse-phase prep-HPLC to afford 102c (82 mg, 13%) as a white solid. MS: [M+H]+ 625.4
