Procedure
Estrone-quinol (100 μM, 286 μg/mL) was incubated at 37° C. in 0.1 M phosphate buffer (pH 7.5) in the presence of NADPH (1 mM) and male rat liver microsomes (1 mg/ml final protein concentration). Aliquots (500 μl) were taken after 5 minutes of incubation with rat liver microsomes. Estrone quinol was found to undergo reduction to form estrone. In addition, the concentration of estrone reached after 5 minutes of microsomal incubation was about 12-times higher (15.1 μg/mL) than the value of 1.2 μg/mL measured without the addition of the microsomes in the relevant control experiment (NADPH present but microsomes not added.
