A solution of 5-[4-({N-FMOC-O-t-butoxycarbonylseryl}amino)phenoxy]-2-amino-1H-indole-3-carbonitrile (1.45 g; 2.3 mmol) in CHCl3, (30 ml) was treated dropwise with piperidine (3.25 ml). The mixture was stirred for 3 hours at room temperature and was directly purified by flash chromatography eluting with increasingly polar mixtures of MeOH/CH2Cl2 (2.5 to 10% MeOH). The appropriate fractions were evaporated to give 5-[4-(O-t-butoxycarbonylserylamino)phenoxy]-2-amino-1H-indole-3-carbonitrile as a white solid (878 mg).
