2-Amino-5-[2,6-difluoro-4-(1-hydroxy-1-methylethyl)phenyl]thiophene-3-carboxamide (90 mg, 0.29 mmol), 2-chloro-6-(2-hydroxyethyl)-6,7-dihydro-5H-pyrrolo[3,4-b]pyridin-5-one (61 mg, 0.29 mmol), Pd2dba3 (26.4 mg, 0.029 mmol), K2CO3 (43.8 mg, 0.317 mmol) and X-Phos (68.7 mg, 0.144 mmol) were added to a 2 mL microwave vial. Degassed tert-amyl alcohol (0.7 mL) was added and the vial evacuated and back-filled with nitrogen (3×). The resulting mixture was stirred at 100° C. overnight. Room temperature was attained, MeOH was added and the resulting mixture filtered through Celite. The filtrate was concentrated in vacuo while loading onto silica. Purification of the residue by silica gel chromatography (0-20% MeOH/EtOAc) gave the title compound as a pale yellow solid after triturating in EtOH. LRMS (APCI) calc'd for (C23H23F2N4O4S) [M+H]+, 489. found 489.
