Procedure

Bromine (1.9 g, 12 mmol, 1.2 equiv.) was added dropwise to a stirred solution of 3-benzyl-2-propylquinazolin-4(3H)-one (1–2, 2.78 g, 10.0 mmol, 1 equiv.) and sodium acetate(0.98 g, 12 mmol, 1.2 equiv.) in acetic acid (10 mL). The reaction was warmed at 45° C. for 2 hours and then poured into water (300 mL). The resulting mixture was stirred for 2 hours and the solid collected by filtration to provide 3-benzyl-2(1-bromopropyl)quinazolin-4(3H)-one (1–3) as an off-white solid. 1H NMR (500 MHz, CDCl3)δ8.35 (d, J=7 Hz, 1H), 7.64 (m, 2H), 7.53 (t, J=7 Hz, 1H), 7.33 (m, 2H), 7.28 (m, 1H), 7.15 (d, J=7 Hz, 2H), 6.22 (d, J=16 Hz, 1H), 4.95 (d, J=16 Hz, 1H), 4.64 (t, J=7 Hz, 1H), 2.48 (m, 1H), 2.25 (m, 1H), 0.77 (t, J=7 Hz, 3H).
