Reaction Steps

A mixture of (intermediate 26) (0.04 mol) in DMF (200 ml) was cooled to 5° C. under N2 flow. NaH 80% in oil (0.048 mol) was added portionwise. The mixture was stirred at 5° C. for 30 min. Then iodomethane (0.048 mol) was added dropwise. The mixture was stirred at 5° C. for 30 min, hydrolized and poured out into water. The precipitate was filtered off, washed with water and taken up in CH2Cl2. The organic layer was separated, dried (MgSO4), filtered and the solvent was evaporated till dryness. The residue (15.5 g) was purified by column chromatography over silica gel (eluent: CH2Cl2/EtOAc 85/15; 15–40 μm). The pure fractions were collected and the solvent was evaporated. The residue (13.3 g, 91.7%) was crystallized from CH3CN and DIPE. The precipitate was filtered off and dried, yielding 1.5 g (9.6%) of 4-(3-chlorophenyl)-6-(3-furanylcarbonyl)-1-methyl-2(1H)-quinolinone (intermediate 27), melting point 192° C.
