Potassium permanganate (0.335 g) and 18 crown-6 (10 mg) were added to a solution of 2-[1-hydroxy-1-(1-naphthalenyl)methyl]-5-methyl-7-(2methylpropyl)thieno[2,3-d]pyridazin-4(5H)-one (example 7, 0.40 g) in dichloromethane (30 ml) at room temperature. After 1 hour, further potassium permanganate (0.30 g) was added and stirring was continued for a further hour. The mixture was filtered, diluted with dichloromethane (70 ml), washed twice with water then with brine, then dried, filtered and evaporated. The residue was purified by column chromatography, eluting with ethyl acetate:isohexane (1:2), to give the title compound (0.14 g). This was purified further by preparative normal-phase HPLC with gradient ethyl acetate/isohexane elution.
