Reaction Steps
To a suspension of pyridin-2-yl-acetic acid hydrochloride (60 mg in 0.8 mL methylene chloride) was added 0.051 mL triethylamine and the mixture stirred at room temperature for 10 minutes. At this time, 98 mg 1-(3-dimethylaminopropyl)-3-ethylcarbodiimide hydrochloride, 69 mg 1-hydroxybenzotriazole, and 60 mg 2-{2-[6-amino-7-chloro-3-(3,5-dimethylphenyl)-2-oxo-1,2-dihydro-quinolin-4-yloxy]-ethyl}-piperidine-1-carboxylic acid tert-butyl ester (prepared essentially as described in Example 1) were added in order. After 22 hours, the mixture was diluted with methylene chloride and extracted sequentially with water, 10% citric acid and brine. The organic portion was dried over sodium sulfate and the concentrate purified by flash chromatography on silica gel (hexane:ethyl acetate, 1:1; then methylene chloride:10% ammonium hydroxide in methanol, 95:5) to give the title compound, 55 mg.
