Reaction Steps
To a solution piperazin-3-one-1-carboxylic acid tert-butyl ester (2.11 g, 10.5 mmol) in dry DMEF (20 mL) at 0° C. was added sodium hydride (60% dispersion in mineral oil, 632 mg, 15.8 mmol). The reaction mixture was stirred for 1 hour, and then a solution of the mesylate from Step C (5.47 g, 10.5 mmol) in DMF (10 mL) was added. The reaction mixture was stirred for 16 hours. The solvent was removed in vacuo and the residue partitioned between methylene chloride (100 mL) and saturated aqueous sodium bicarbonate (100 mL). The layers were separated and the aqueous layer was extracted with methylene chloride (3×100 mL). The combined organic layers were dried over sodium sulfate, filtered, and concentrated in vacuo. The crude product was purified by column chromatography (20→50% ethyl acetate/hexane) to provide the title compound as a yellow oil.
