Procedure

All isomers of PGF2α were dissolved or diluted in adequate amount of ethanol and after aliquot, stored at −70° C. As an internal standard, 10 ng of 8-iso-PGF2α-d4 was mixed with 0.5 ml of plasma in a glass tube and was then purified by a C18 Solid-Phase Extraction column (J. T. Baker, M A, USA). Sample eluants were evaporated under a stream of nitrogen and re-dissolved in 50 μl acetonitrile:water (15:85 v/v) solution followed by vortex for 30 seconds and 10 μl extract was further analyzed with a LC/MS/MS system.
