Reaction Steps
To a solution of 4-[(3-hydroxypropyl)amino]-7-(trifluoromethyl)imidazo[1,2-a]quinoxalin-1-carbaldehyde (35 mg, 0.10 mmol) in methanol (1.2 mL) was added glyoxal (40% in water, 15 μL, 0.10 mmol). The mixture was cooled at 0° C. and NH3 (2M in ethanol, 150 μL, 0.30 mmol) was added. The reaction was stirred at 25° C. under argon overnight. The reaction was followed by TLC that showed small conversion of the starting material in imidazole. The addition of reactants was repeated 4 times in 4 days, until the TLC indicated completion of the reaction. The reaction was concentrated. The crude product was purified twice by preparative TLC (dichloromethane/methanol 9/1) to give the title compound (7 mg, 18%) as a beige powder.
