Procedure

Step-5: A solution of benzyl 3-((2-amino-3-carbamoylphenyl)carbamoyl)-3-methyl-3,4-dihydroisoquinoline-2(1H)-carboxylate (40 mg, 0.087 mmol) in acetic acid (2 ml) was heated to 80° C. and stirred overnight. The reaction was further heated to 120° C. for 3 h. The reaction mixture was concentrated, neutralized with aq. NaHCO3 and extracted with ethyl acetate. The ethyl acetate layer was washed with brine, dried over anhydrous sodium sulfate and evaporated. Purification of the crude product by preparative TLC using hexane/ethyl acetate (1:1) mixture as eluent afforded benzyl 3-(4-carbamoyl-1H-benzo[d]imidazol-2-yl)-3-methyl-3,4-dihydroisoquinoline-2(1H)-carboxylate. MS (ES+): m/z 441.22 [M+H]+
