Procedure
To a suspension of benzyl trans-(±)-3-{[2-(1-oxido-2,3-dihydro-1,4-benzothiazepin-4(5H)-yl)-6-methylthienopyrimidin-4-yl]amino}-4-fluoropyrrolidine-1-carboxylate (320 mg, 0.57 mmol) in methanol (5 mL) was added an aqueous solution of potassium hydroxide (40%, 5 mL). The suspension was heated under reflux for 30 minutes. The organic solvent was removed by concentration in vacuo. The residue was purified by preparative HPLC to afford the pure product as a white solid. MS obsd. (ESI+) [(M+H)+] 432, 1H NMR (400 MHz, METHANOL-d4) δ ppm 7.72 (m, 2H), 7.48 (m, 2H), 6.83 (s, 1H), 5.18-5.38 (m, 1H), 4.66-4.99 (m, 5H), 3.26-3.5 (m, 6H), 2.50 (s, 3H).
