Reaction Steps

To a mixture of 2-[1-(4-amino-2-fluoro-phenyl)-piperidin-4-ylidene]-N,N-diethyl-2-(3-fluoro-phenyl)-acetamide (0.33 mmol) and TEA (2.0 mmol) in DCE (10 mL) was added 2-ethylbutyryl chloride (1 mmol) dropwise. The mixture was stirred at rt for 16 h, then was washed with H2O (10 mL). The organic layer was concentrated and the residue was purified by PTLC to provide the title compound (54 mg, 33%). MS (ESI): mass calcd. for C29H37F2N3O2, 497.29. m/z found, 498.4 [M+H]+. 1H NMR (CDCl3): 7.58-7.53 (m, 1H), 7.47 (NH, 1H), 7.37-7.32 (m, 1H), 7.18-7.06 (m, 3H), 7.04-6.97 (m, 1H), 6.92-6.85 (m, 1H), 3.50-3.00 (m, 6H), 2.62-2.52 (m, 4H), 2.10-2.02 (m, 1H), 1.79-1.67 (m, 4H), 1.62-1.52 (m, 2H), 1.16 (t, J=7.1, 3H), 1.00-0.87 (m, 9H).
