Procedure

A round-bottom flask was charged with 3-(benzyloxymethyl)-1H-pyrazol-5-amine (1.74 g, 8.56 mmol, Honma, T. et al. “A Novel Approach for the Development of Selective Cdk4 Inhibitors: Library Design Based on Locations of Cdk4 Specific Amino Acid Residues.” J. Med. Chem. Vol. 44, No. 26 (2001): 4628-4640), sodium nitromalonaldehyde monohydrate (1.46 g, 9.42 mmol) and acetic acid (8.7 mL). The reaction mixture was heated at 100° C. for 24 hours. The reaction mixture was poured into water and then extracted with ethyl acetate (2×). The combined organic layers were dried with sodium sulfate, filtered and concentrated in vacuo. The crude product was purified by flash chromatography to afford 3-(benzyloxymethyl)-5-nitro-1H-pyrazolo[3,4-b]pyridine (1.37 g, 56%) as a solid.
