A reaction mixture of 4-(2-amino-pyrimidin-5-yl)-N1-(1,1-dimethyl-propyl)-benzene-1,2-diamine (130 mg, 0.48 mmol), 2-1,2,4-triazol-1-yl-benzaldehyde (110 mg, 0.64 mmol) and L-proline (5 mg) in MeOH (25 mL) is refluxed for 16 hours. The reaction mixture is allowed to cool to room temperature and is concentrated under reduced pressure. The residue is purified by silica gel flash column chromatography with 8% MeOH in CH2Cl2 as the eluent to afford 5-[1-(1,1-dimethyl-propyl)-2-(2-1,2,4-triazol-1-yl-phenyl)-1H-benzimidazol-5-yl]-pyrimidin-2-ylamine. LCMS (ESMS): m/z 425.20 (M++1)
