Reaction Steps

To a solution of {4-amino-5-[3-(benzyloxy)phenyl]pyrrolo[2,1-f][1,2,4]triazin-7-yl}methanol (300 mg, 0.87 mmol) in dichloromethane (5 mL) was added Dess-Martin Periodinane (492 mg, 1.13 mmol). The mixture was stirred (rt) for 4 h. The reaction was quenched with the addition of a 1:1 mixture of saturated, aqueous NaHCO3 and saturated aqueous Na2S2O3 solutions (5 mL) and the mixture was allowed to stir at it for 1 h. The mixture was extracted with dichloromethane (3×5 mL) and the combined organics were washed with brine, dried (Na2SO4) and concentrated to afford 105 mg (35%) of the desired product, which was used without further purification. ES-MS m/z 345.2 [M+H]+, HPLC RT (min) 3.12.
