Reaction Steps
Add K2CO3 (220 mg, 1.6 mmol) to a solution of 4-cyano-5-ethyl-3-(2′-hydroxy-biphenyl-4-yl)-thiophene-2-carboxylic acid ethyl ester (200 mg, 0.53 mmol) in 2 mL of dry DMF at 0° C. and stir at this temperature for 1 hour. Treat with n-propyl iodide (0.2 mL, 2 mmol) and allow to warm to room temperature. After 3 hours pour into 25 mL cold 0.2N HCl. Extract with EtOAc (2×50 mL) and wash the combined EtOAc layers with water (2×20 mL) and brine (1×20 mL). Dry over Na2SO4, filter and evaporate to an oil. Chromatograph on silica gel (1/9-1/3 EtOAc/hexanes) to give the title compound as a tan solid, 158 mg (71%). HPLC=99%.
