Reaction Steps

To a solution of 3-((R)-3-((S)-1-(4-bromophenyl)ethyl)-6-(4-fluorophenyl)-2-oxo-1,3-oxazinan-6-yl)propanal (300 mg, 0.69 mmol) in THF (4 ml) was added dropwise MeMgBr (0.7 ml, 3 mol/L) at −78° C. The mixture was stirred for 2 h and the temperature was allowed to be warm to rt at this stage. The reaction was quenched with aqueous NH4Cl. The organic layer was separated, and the aqueous layer was extracted with EtOAc. The combined organic layer was washed with brine, dried over Na2SO4 and concentrated to give the crude product, which was purified by HPLC and chiral HPLC to give two isomers.
