To a mixture of N6-(2-isopropoxy-5-methyl-4-(piperidin-4-yl)phenyl)-N4-(2-(isopropylsulfonyl)phenyl)-3-methyl-1H-pyrazolo[3,4-d]pyrimidine-4,6-diamine (50 mg, 0.087 mmol) in CH2Cl2 (3 mL) was added bromoacetyl chloride (10 uL, 0.095 mmol) dropwise at 0° C. The solvent was removed under reduced pressure and the residue was purified by silica gel chromatography, eluting with a gradient of EtOAc/hexane (0-100%) to give the title compound (2-bromo-1-(4-(5-isopropoxy-4-(4-(2-(isopropylsulfonyl)phenylamino)-3-methyl-1H-pyrazolo[3,4-d]pyrimidin-6-ylamino)-2-methylphenyl)piperidin-1-yl)ethanone) as a light yellow solid. MS: m/z=698.2 (M+1).
