Phenylmethyl {(2S)-3-{[5-chloro-4-(4-chloro-1-methyl-1H-pyrazol-5-yl)-2-thienyl]amino}-2-[(3,4-difluorophenyl)methyl]-3-oxopropyl}carbamate (91 mg, 0.157 mmol) was dissolved in a solution of thioanisole (0.286 ml) in trifluoroacetic acid (2.855 ml). After 4 h at 25° C., the reaction mixture was dry loaded onto silica and neutralized/purified through silica chromatography (0-15% MeOH in DCM (1% NH4OH)) affording the free base of the title compound.
