Reaction Steps

To (S)-tert-butyl 3-(2-(4-(3-ethylureido)phenyl)-9-methyl-6-(3-methyl-morpholino)-9H-purin-8-yl)-3-hydroxyazetidine-1-carboxylate (45.2 mg, 0.08 mmol) in anhydrous dichloromethane (3.0 mL) and anhydrous methanol (3.0 mL) at 0° C. was added 4M hydrogen chloride in 1,4-dioxane (0.20 mL, 0.80 mmol, 10.0 eq.). The reaction mixture was stirred at RT under N2 for 16 h. Trifluoroacetic acid (0.20 mL) was added and the reaction mixture was stirred at 40° C. under N2 for 24 h. The reaction was then cooled to RT. The reaction mixture was concentrated in vacuo, and the residue was diluted with ethyl acetate. The organic layer was washed with saturated aqueous solution of sodium bicarbonate, water and brine, dried (Na2SO4), filtered and concentrated in vacuo. The crude was purified by reverse phase HPLC to give the title compound as a white solid (7.8 mg, 21%). 1H NMR (DMSO-d6, 400 MHz) δ ppm 8.70 (s, 1H), 8.27 (d, J=8.8 Hz, 2H), 7.95 (s, 1H), 7.48 (d, J=8.8 Hz, 2H), 6.44 (s, 1H), 6.22 (t, J=5.5 Hz, 1H), 5.48 (broad s, 1H), 5.15 (broad s, 1H), 4.14 (d, J=8.2 Hz, 1H), 4.08 (d, J=8.3 Hz, 1H), 4.01 (d, J=8.3 Hz, 1H), 3.84 to 3.67 (m, 7H), 3.61 to 3.52 (m, 1H), 3.50 to 3.39 (m, 1H), 3.17 to 3.07 (m, 2H), 1.34 (d, J=6.7 Hz, 3H), 1.06 (t, J=7.2 Hz, 3H); LC-MS m/z (method C)=467.2 [M+H]+, RT=3.21 min.
