Reaction Steps

To a solution of 5-bromo-N-(4-fluoro-3-methoxyphenyl)pyrimidin-2-amine 44 (0.25 mmol) in 3 mL of anhydrous DCM at 0° C. is added BBr3 (1.27 mmol) drop wise. The reaction mixture is stirred at rt for 12 h. After this time the reaction mixture is diluted with a 2M solution of Na2CO3 and extracted with DCM (2×20 mL). The organic layer is washed with brine, dried over Na2SO4 and concentrated to afford 5-(5-bromopyrimidin-2-ylamino)-2-fluorophenol 45 as brownish solid which is used without further purification. MS (m/z) (M+1)+: 286.2.
