To a stirred solution of tert-butyl 4-(2-oxopropyl)pyridin-2-ylcarbamate (46) (300 mg, 1.20 mmol) in methanol (12 mL) at 0° C., under nitrogen, was added sodium borohydride (45.3 mg, 1.20 mmol). After 10 min the reaction mixture was warmed to RT and stirring continued for 75 min. The mixture was evaporated in vacuo and the residue was taken up into EtOAc and washed with aqueous NaHCO3 solution. The aqueous layer was extracted twice with EtOAc and the combined organic extracts were washed with brine, then dried (MgSO4) and evaporated in vacuo. The residue was purified by flash column chromatography (SiO2, 12 g, 10-80% EtOAc in isohexane, gradient elution) to afford the title compound (47) (262 mg, 85%) as a white powder; m/z 253.0 (M+H)+ (ES+).
