Reaction Steps

To a dimethylformamide solution (200 ml) of the above Compound 6 (3.56 g, 11.4 mmol) was added N-iodo succinimide (3.06 g, 13.6 mmol), and the solution was stirred overnight at room temperature. The reaction solution was distilled off under reduced pressure, and the residue was added with chloroform, washed with 10% sodium hydrogen sulfite aqueous solution and water, and dried over anhydrous sodium sulfate, and then the solvent was distilled off. Precipitated crystals were collected by filtering, and washed with methanol to give 3-(4-fluorobenzyl)-8-hydroxy-5-iodo[1,6]naphthyridine-7-carboxylic acid methyl ester 7 (4.97 g) in 99% yield.
