Reaction Steps
A mixture of 1-(4-chlorobutyl)-3-(methyloxy)-2(1H)-pyridinone (67.8 mg, 0.31 mmol), 2-methyl-5-(1-piperazinyl)quinoline (see WO2004/046124 D3) (77.5 mg, 0.34 mmol), triethylamine (0.09 mL, 0.64 mmol) and a catalytic amount of sodium iodide in anhydrous N,N-dimethylformamide (2.50 mL) was stirred at 110° C. overnight. The reaction mixture was concentrated in vacuo, taken-up in dichloromethane (5 mL) and washed with water (2×5 mL). The organic layer was dried (Na2SO4) and evaporated in vacuo. The crude product was passed through a SCX cartridge, washing with methanol and eluting with ammonia 2M in methanol and then purified by flash chromatography on silica gel, eluting with 2% methanol in dichloromethane, to afford the corresponding free base of the title compound as a yellow solid (65.9 mg, 52%).
