Reaction Steps
Tetrakis(triphenylphosphine)palladium(0) (13 mg, 0.011 mmol) was added to a stirred mixture of (1R*,2R*,5S*,7S*)-2-{[3,5-bis(trifluoromethyl)phenyl]methoxy}-7-cyano-1-phenyl-8-(prop-2-enyl)-8-azabicyclo[3.2.1]octane (Example 57; 68 mg, 0.14 mmol), 1,3-dimethylbarbituric acid (70 mg, 0.5 mmol) in dichloromethane (3 ml) at room temperature. The reaction mixture was stirred at 30-35° C. for 1 hour and treated with 2M aqueous NaOH (5 ml). The mixture was stirred for 5 minutes, then diluted with water (30 ml) and extracted into dichloromethane. The combined organic extracts were dried (Na2SO4) and concentrated. The residue was purified by chromatography on silica gel to give the title compound (37 mg, 60%). The hydrochloride salt of the title compound was prepared by treatment with ethereal HCl.
