Reaction Steps
To a solution of 3-methoxy-2-naphthoic acid (Method A6, Step 2; 0.762 g, 3.80 mmol) and Et3N (0.588 mL, 4.2 mmol) in anh toluene (20 mL) at room temp. was added a solution of diphenylphosphoryl azide (1.16 g, 4.2 mmol) in toluene (5 mL). The resulting mixture was heated to 80° C. for 2 h, cooled to room temp., and p-toluidine (0.455 g, 4.1 mmol) was added. The mixture was heated at 80° C. overnight, cooled to room temp., quenched with a 10% citric acid solution, and extracted with EtOAc (2×25 mL). The combined organic layers were washed with a saturated NaCl solution (25 mL), dried (MgSO4), and concentrated in vacuo. The residue was triturated with CH2Cl2 to give the desired urea as white powder (0.700 g, 61%): mp 171-172° C; 1H-NMR (DMSO-d6) δ 2.22 (s, 3H), 3.99 (s, 3H), 7.07 (d, J=8.49 Hz, 2H), 7.27-7.36 (m, 5H), 7.67-7.72 (m, 2H), 8.43 (s, 1H), 8.57 (s, 1H), 9.33 (s, 1H); FAB-MS m/z 307 ((M+H)+).
