Potassium tert-butoxide (21 mg, 0.19 mmol) was added to a solution of 2-[4-(2,2-dimethylbutyl)-1-trityl-1H-imidazol-2-yl]-1-[4-(5-fluoropyridin-2-yl)phenyl]ethanol (104 gm, 0.17 mmol) and methyl iodide (19 μL, 0.31 mmol) in tert-butanol (17 mL) at ambient temperature. After stirring overnight, the reaction mixture was washed with brine (100 mL), dried (magnesium sulfate) and concentrated in vacuo. Chromatography over silica eluting with 0-50% ethyl acetate/hexane afforded 2-(4-{2-[4-(2,2-dimethylbutyl)-1-trityl-1H-imidazol-2-yl]-1-methoxyethyl}phenyl)-5-fluoropyridine as an off-white solid.
